I have a very similar system (MB: ASRock Z97 Extreme6, BIOS: v2.50, System drive: Samsung 950 Pro SSD connected to M.2_1 port, OS: Win10 x64 installed in "clean" UEFI mode) and can confirm your report.

It is impossible to successfully cold boot into Win10, when the OS has been installed onto a Samsung 950 Pro SSD.
Each time, when I power on my computer after having it completely powered off for a while, the boot procedure hangs. The circle stands still and nothing happens anymore (no BSOD and no error message). The SSD is running fine and will boot instantly after having turned off and turned on the computer after a few seconds. The issue doesn't happen while rebooting, only while doing a cold boot.
Since I have done a lot of tests with my ASRock Z97 Extreme6 running different PCIe and M.2 connected SSDs (look here: http://www.win-raid.com/t977f34-Benchmarks-Comparison-of-the-currently-fastest-consumer-SSDs.html), I am pretty sure, that it is just the combination of the ASRock Z97 Extreme6 mainboard with the Samsung 950 Pro SSD, which has this issue. All other M.2_1 connected SSDs worked fine even while doing a cold boot.
I suspect, that the problem is caused by a misunderstanding between the NVMe related modules of the mainboard BIOS and the Firmware of the Samsung 950 Pro SSD.
